Abstract


DNA SEQUENCE ANALYSIS OF NATIVE TRICHODERMA LONGIBRACHIATUM AND PRODUCTION OF BIOTECHNOLOGICAL FUNGAL SILVER NANOPARTICLE (AGNP)

Nanopartices, which are known as nanotechnological materials, are also considered as biotechnological products. Biological methods are more economic compare to chemical and physical methods in R&D studies. Production of nanoparticles (AgNPs) by biotechnological processes are faster than other processes. Trichoderma sp. produces a wide variety of extracellular industrial enzymes and metabolites. In the present research we report our optimisation experimental study of the extracellular synthesis of silver nanoparticles using Trichoderma longibrachiatum. Biological silver nanoparticle was synthesized extracellularly by using the fungus. This study contains macroscopic, microscopic and molecular characterization of Trichoderma sp. Molecular identification of 18s rDNA and ITS regions by PCR amplification was provided by using Fungus-specific primers. The most suitable temperature for AgNP biosynthesis was 55 ?, substrate concentration was 1:100 and pH was 12. Antibacterial effect of AgNPs was determined by agar diffusion method. At the end of the experiments, it was determined that the biosynthesis of AgNP showed antimicrobial activity against E. coli, S. aureus, B. subtilis. At the end of the experiments, it was determined that the biosynthesis of AgNP showed antimicrobial activity against Escherichia coli, Staphylococcus aureus, Bacillus subtilis.
